چکیده

Chronic, excessive exposure to solar ultraviolet radiation is the major causative agent for cutaneous squamous cell carcinomas. Mutations in p53 are essential these cancers develop. UV also immunosuppressive and an additional requirement UV-induced SCCs can be reversed with compound CP31398, which results fewer mice exposed a skin carcinogenesis protocol. It unknown whether mutations contribute photoimmunosuppression. The purpose of this study was determine if effects could by pretreatment CP31398. A local UVB regimen consisting (200 mJ/cm2) 4 days followed sensitization hapten 2, 4, dinitrofluorobenzene (DNFB) employed To role CP-31398, we treated shaved dorsal C57BL/6 CP-31398 (1.25 mg/mouse) or vehicle cream, 30 min before each treatment. Treatment did not abrogate effect UV. These findings suggest that although responsible SCCs, they necessary immunosuppression.

Prostaglandin E2 Promotes UV radiation-induced immune suppression through DNA hypermethylation.

Exposure of mice to UV radiation results in suppression of the contact hypersensitivity (CHS) response. Here, we report that the UV-induced suppression of CHS is associated with increases in the levels of cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), and PGE2 receptors in the exposed skin. UV radiation-induced suppression of CHS was inhibited by topical treatment of the skin with celecoxib...

Mechanisms underlying UV-induced immune suppression: implications for sunscreen design.

The ultraviolet (UV) radiation present in sunlight is immune-suppressive. Recently we showed that solar-simulated UV radiation (UVA + UVB; 295-400 nm), applied after immunization, suppressed immunological memory and the elicitation of delayed-type hypersensitivity to the common opportunistic pathogen, Candida albicans. Further, we found that wavelengths in the UVA region of the solar spectrum (...

Latent expression of p53 mutations and radiation-induced mammary cancer.

EF42 is a clonally derived preneoplastic cell lineage from irradiated mouse mammary tissue, which becomes neoplastic with time in vitro or in vivo. We now report that multiple mutations in p53 occur before the acquisition of the neoplastic phenotype. The selective expansion of mutant cells is accompanied by loss of heterozygosity at the p53 locus and c-myc amplification. Although p53 mutations ...

Ultra-wide band electromagnetic radiation does not affect UV-induced recombination and mutagenesis in yeast.

Cell samples of the yeast Saccharomyces cerevisiae were exposed to 100 J/m2 of 254 nm ultraviolet (UV) radiation followed by a 30 min treatment with ultra-wide band (UWB) electromagnetic pulses. The UWB pulses (101-104 kV/m, 1.0 ns width, 165 ps rise time) were applied at the repetition rates of 0 Hz (sham), 16 Hz, or 600 Hz. The effect of exposures was evaluated from the colony-forming ability...

Radiation-induced mutations at the autosomal thymidine kinase locus are not elevated in p53-null cells.

To explore further the possibility that some forms of mutated p53 may increase mutagenesis in a positive manner, a double p53 knockout cell line was created, using a promoterless gene targeting approach. The identity of these p53-null cells was confirmed by Southern blot and Western blot analyses. Radiation-induced toxicity and mutagenicity was then compared among p53-null cells, TK6 cells with...
